The preparation process of strong fluorescent phycocyanin using fresh smashed algae as raw material includes: washing with EDTA containing phosphoric acid buffering solution, ultrasonic crushing of cell to release pigment protein, centrifugal separation to eliminate insoluble cell membrane and organelle and to collect the supernatant; salting out phycocyanin precipitant; dissolving the precipitant in EDTA containing phosphoric acid buffering solution, desalting with Sephadex G-25, eliminating alkali impurity protein in DEAE-Sepharose FF column; obtaining phycocyanin eluent in hydroxyapatite column; eliminating acid impurity protein in CM-Sepharose FF column; and finally obtaining high-purity strong fluorescent phycocyanin in hydroxyapatite column. The process is suitable for large-scale industrial production.
